There not lesbians or Man haters, just Vampires and as such (combined with the Transhuman Treachery trope) they no longer see them selves as humans on a fundimental level and makes them kinda psycopathic (It's easier be a immortal blood sucking creature of the night if you no longer see humans as people or if you see your self as a superior being).

Lori may not be a lesbian. I've known quite a few lesbians in my time and there were only one or two that had any antipathy toward my gender. But Lori's response upon seeing Leonard in her outfit ("No amount of dry-cleaning will get that man-stench out .") shows a disgust towards men in general. I was rooting for her to defeat the gorgon, Bianca. Now I see I was cheering for the wrong side. Hopefully Miss Cresida can get her revenge in the future, and any others of her family as well, by turning the Bakertown cheerleaders into a buffet. Mwa, ha, ha, ha...

(But in all seriousness if you were hoping for a "nice" vampire protagonist then you're looking for something like Karin/Chibi Vampire or Rosario + Vampire)

Inner Moka may not be conventionally nice, but she is at least honourable. Kahlua would be a sweetheart if her dad hadn't made her the family assassin, obliging her to kill. Aqua... I can't find anything nice to say about Aqua. She's psycho.
